ISOLATION AND CHARACTERISTICS OF BACTERIA CITROBACTER SP. SR35 FROM A COAL MINE WASTE DUMP

SummaryBacteria of the genus Citrobacter are found in soil, water, intestinal tract animals, human clinical samples (urine, sputum, blood, wound drainage, etc.), also wastewaters and waste dumps mines. Gram-negative bacteria SR35, capable reducing sulfur sulfate ions, were isolated from dump Nadiya coal mine, Chervonograd Mining Industrial district (Lviv region, Ukraine). The work aimed to identify study morphological, physical chemical properties (cell shape, size, Gram staining, sporulation, motility, oxygen requirements, ability form H2S, utilization carbon sources, catalase activity, oxidase activity) isolate SR35. Methods. We used standard microbiological biochemical research methods (microbial culture, microscopy methods, determination amilase, lipase activity). Chromosomal DNA was by method soft lysis. 16S rRNA gene amplified using universal primers 27F 1492R. It sequenced Sanger method. Phylogenetic reconstruction performed MEGA X program. Identification isolates carried out based on both sequence physiological properties. Results. studied rods (0.5–0.8×1.5–2.0μm), which chains nitrate ions assimilating several sources. Glucose, lactose, mannose, mannitol, inositol fermented with formation acid. Motile. Catalase positive, negative. Form H2S during growth Kligler’s medium. According results pairwise alignment isolate’s sequence, highest percentage identity representatives established (99.23–99.86% identity, coverage 98%) it confirmed phylogenetic reconstruction. sp. SR35 is resistant 2 μM cadmium (II), 5 mM iron 0.25 cobalt 10 manganese 0.5 copper 0.1 chromium (VI). Conclusions. sequencing (accession number GenBank OP279754) gene, physiological, characteristics (oxidase, catalase, metabolism production that belongs Citrobacter.
